Using the keyboard
To save as PDF (PNG as of Tiger) on your Desktop:
- ⌘-Shift-3: Full screen shot
- ⌘-Shift-4: Select the area you want to capture
- ⌘-Shift-4, then Space: Select the window on the screen for capture and click on it.
To copy the screen content to the clipboard:
- ⌘-Shift-Ctrl-3: Full screen;
- ⌘-Shift-Ctrl-4: Area;
- ⌘-Shift-Ctrl-4, then Space: Window.
For pre-Tiger users only:
Once you have a PDF, you can use Preview to Export to most any image format. If you capture to the clipboard, you can simply press ⌘-N in Preview to create a new file with your screenshot, which you can export to png or whatever without the intermediary PDF.

Notes
When you take a window screenshot with ⌘-Shift-4+Space, it's taken before compositing, so if it's a transparent window, the window(s) behind it will not show through.
